LATE PCR Partnership programme

LATE PCR expands the boundaries of what is possible with PCR. Bio-Seeq takes that capability out of the laboratory and opens up the possibility of powerful ‘near patient’ or ‘in the field’ diagnostics.

System configuration

The HI-TIP system provides a library divided into categories and subcategories containing combined, complete threat images and images of individual threat objects. The supervisor can add, edit and delete categories.

Moreover, users with supervisor access level can specify:


  • from which categories and at which percentage in proportion to the other categories combined threat images and single threat objects are projected during the normal operational flow
  • which images from a category are to be projected
  • which decision times are provided for the operator to detect a threat image
  • how many projections are made randomly rather than within the diversification range
  • in what time the operator should detect a threat image
  • the number of projections compared with the number of real bags
  • the ratio between fictional projection and number of real bags

The Mail & parcels section includes products especially for the non-intrusive inspection of smaller items, i.e. mail and parcels. One applicable selection criterion is the maximum object size in the inspection process. All X-ray systems in this category are based on the proven SiProX and HiTraX technology, offering superior image quality and excellent usability.

Millimetre-waves lie in the spectral region between radio waves and infrared. This band possesses a unique property of passing transparently through lightweight materials such as clothing fabrics. The property is made use of in mm-wave imagers that find their principal application in concealed object detection systems – typically used to identify the presence of weapons and contraband that are hidden beneath a persons clothing.

Millimetre-wave imagers have evolved over the past 50 years from very rudimentary systems to today’s technologies that create live video-like images for use as advanced people screening portals. In this evolution from scientific instruments to commercial security devices, the overriding constraint has been the cost of the sensors that detect the mm-wave signals that are needed to produce an image. To overcome this barrier, scanning systems have been employed to direct mm-wave energy from all parts of the scene onto a small number of sensors. Historically these scanning systems have been based on mechanical techniques employing moving parts to direct and focus the mm-wave energy. Smiths use a revolutionary approach that scans the scene electronically and has resulted in the first ‘no moving parts’ imager that is suitable for mass people screening.

The eqo product – named because it senses mm-wave echoes – marks an evolution to ‘next-generation’ technology for mm-wave imagers. The electronic scanning not only dispenses with the need for moving parts but also dramatically reduces the footprint of the product. The system consists of a flat-panel array that produces excellent image quality with a real-time, full motion video style output. Heimann Systems

Heimann Systems was founded in 1946 by Prof. Dr. Walter Heimann, one of television technology's early pioneers. 

In the early 1970s Heimann diversified into markets for security technology. Completely new product lines were developed, among them X-ray inspection units for baggage screening which became an international requirement in the 70s & early 80s when aircraft hijacking became a global menace.

In response to the requirements of the 1990s, systems were developed for truck and container inspection and for the automatic detection of explosives in baggage. 

Today more than 40,000 Smiths Heimann X-ray inspection systems have been supplied to more than 150 countries, screening mail, baggage, merchandise, containers and trucks. Heimann regularly exports around 80% of its products and earned a worldwide reputation for product quality, which has its roots in the in-house development department, with experienced scientists and engineers covering the entire R&D spectrum, from basic development through to the integration of complex systems.

When Heimann Systems joined Smiths in December 2002, it virtally doubled the size of the Detection business.

Sample Preparation Unit (SPU)

Field based PCR has been slow in it's adoption, partly due to the requirement for highly purified nucleic acid (DNA or RNA) from samples prior to PCR analysis. The SPU from Smiths Detection is a fully automated sample preparation system designed from the ground up, with field use by the non expert in mind. The SPU is a single use consumable which is capable of purifying DNA or RNA from a wide range of sample types:
• Blood
• Milk
• Vesicular Tissue
• Faeces
• Swabs

The SPU is used in conjunction with the reagent pack. The SPU contains all the mechanical and chemical parts needed to purify either DNA or RNA from a sample. Assay specific reagents are then automatically added, by the SPU, from the reagent pack to the nucleic acid prior to PCR. All steps in the process are handled by the SPU and the user is not required to perform any actions during this time.

Ease of use has been a priority during the development of this system. Samples are placed into the device and the cap screwed in place. The SPU is then placed on the Portable Veterinary Laboratory instrument which will then process the sample and finally run PCR.
 
Reagent Pack
The regent pack is used in conjunction with the SPU. The reagent pack contains the assay specific reagents and is designed to be plugged into a SPU unit just prior to use. This means that the SPU and reagent pack can be bought separately, as needed, reducing the likelihood of having assays on the shelves which are never used or worse still have passed their expiry date when they are needed.

Smiths Detection's reagent pack contains a number of unique features aimed at achieving our goal of simple, reliable operation in harsh field environments. Features such as barcodes, allow the assay to be automatically selected by the instrument rather than the user manually selecting the correct test from a list. Expiration dates can be checked to ensure the assays will work reliably.

Separating the assay specific reagents from the sample preparation module provides a number of benefits to the field user. The reagent kit is much smaller than the SPU and therefore the full range of assays can be carried at all times, ensuring the veterinarian always has the right assay without having to carry the equivalent number of SPUs.

X-ACT principle

X-ACT is based on the proven HI-MAT PLUS multi energy material discrimination X-ray technique, sophisticated image processing software and dedicated hardware.
The HI-MAT PLUS material detection system has been further improved using a highly stabilized X-ray source and selected components for detection. The detection system provides the required material resolution to distinguish between explosives, narcotics and other materials.

The automatic detection software which is run on a state-of-the-art computer hardware is a spin-off of the renowned EDS explosive detection system for hold baggage screening.

The main feature is to separate materials of interest from other bag contents. Even if suspect items are obscured or overlap with others, they can be separated and alerted.
Besides visual framing on the display, the operative can be alerted through acoustic signals or a stopping of the belt.

Customs and border protection requirements are constantly evolving. Traditional fiscal roles continue, such as the collection of excise duties, but there is now additional emphasis on the identification of threats to local and national security – a first line of defence against possible insurgent attacks.
 
The priorities have moved from monitoring cross-border cargo and reducing international shipments of contraband, to screening for explosives, arms, dirty bombs and weapons of mass destruction. Smiths Detection screening systems provide support for all these border control requirements.

Identifying such threats is increasingly more difficult, hidden inside a vehicle or concealed in the middle of the shipment. The challenge is rapid detection without disrupting the daily flow of goods.

Smiths Detection HCV X-ray systems have set the industry standard in cargo inspection for more than 15 years. They are available in various forms, from mobile units ready for use on all terrains in less than 30 minutes, up to fixed high-volume installations. More than 250 cargo screening systems and over 200 mobile cargo systems have now supplied worldwide, in over 50 different countries.

Smiths Detection has a long history of developing simple, rugged detection systems that put sophisticated laboratory technologies into the hands of front line professionals in a wide range of environments. Background to our powerful diagnostics technology Following the 2001 anthrax attacks in the US Smiths Detection developed a portable bio threat detector called Bio-Seeq which used conventional Polymerase Chain Reaction (PCR) technology. In December 2005 the company licensed an advanced form of this technology called LATE (Linear After The Exponential) PCR from Brandeis University in Boston, Mass. We have further developed LATE PCR and the Bio-Seeq platform and added a unique disposable Sample Preparation Unit to create the Bio-Seeq range of nucleic acid detection and analysis systems. The Portable Veterinary Laboratory is moving into production and the Point of Care Diagnostic System will be available early in 2009. The systems provide rapid easy to use tests and meet the needs of Governmental disease screening and outbreak management programmes.

Partnership opportunities
We are keen to make both LATE PCR and our Bio-Seeq platforms widely available and to develop the broadest possible range of assays. To this aim we are developing a LATE PCR Partnership Program and are keen to discuss this with potential collaborators in healthcare, veterinary medicine and other fields.

Clinical Diagnostics - introduction

Molecular diagnostics is widely recognized to be a high growth sector within diagnostics that is having a significant impact on how patients are treated. We have worked closely with industry experts Scientia Advisors (http://www.scientiaadv.com/) to analyse this market. In particular Scientia have helped us to understand the emerging need of healthcare professionals for simple, easy to use molecular diagnostic systems.

We discovered three key un-met needs in this area:
• Simplicity – there is a strong desire for systems that reduce or eliminate manual steps in the sample preparation and analysis process
• Sophistication – most current tests provide simple indications of the presence or absence
of a single organism. There is a need for tests that can identify multiple organisms in a single sample and provide important diagnostic information such as antibiotic resistance or indications
of genetic mutation.
• Speed – a fast time-to result accelerates treatment times and provides important clinical and financial benefits

The Bio-Seeq clinical diagnostics platform is being designed to meet these needs. In particular it will focus on the need for rapid diagnosis of infectious disease in patient screening programmes and in critical care. It will provide simple, easy to use diagnostics suitable for use at or near the Point of Care in Accident and Emergency units, Critical Care laboratories and other locations where speed and accuracy are at a premium.

In addition we seek to: 
• Create solutions for Government sponsored pandemic preparedness programmes.
• Develop field portable diagnostics for use in military medicine
• Establish LATE PCR as a method of testing for genetic markers in the early stages of selected cancers.

Protect buildings against the accidental or deliberate release of chemicals, shutting down HVAC systems. Use intelligent video systems to help in assessing the level of threat and to monitor events.

GID-3 (24/7) - low cost point chemical warfare (CW) detector with minimal through life costs for fixed internal or external sites. Runs for minimum one year without operator intervention.

Classification

In the multi-energy method for evaluation of material information of the X-ray signal, the high and low energy radiation fractions obtained after penetration of the object, are compared. By means of this method conclusions concerning material origins can be made.
The classification of materials is carried out according to the atomic numbers Z. Three main groups of elements are distinguished by colors of a continuous color scale:

0 < Z < 10 orange (low)
10 < Z < 18 green (medium)
18 < Z < 40 blue (high)

Barringer Instruments

Barringer Instruments was founded in 1961 and was initially focused on the geophysical market place for detection and survey instrumentation that helped in the exploration for oil and valuable minerals.  Over the years, Barringer broadened to include instrumentation for nuclear power plants and in the late 1980s turned it attention mainly to narcotics and explosives detection using Ion Mobility Spectrometry (IMS). 

During the 1990s Barringer became a world leader in trace detection instruments for drugs and explosives detection, providing equipment to over 50 countries with thousands of units deployed worldwide.  In 2000, Barringer began marketing IMS technology to the Life Sciences market and pharmaceutical manufacturers, offering a faster and more accurate process for cleaning validation, raw material identification, quality control and dissolution testing. 

In April 2001 Smiths acquired Barringer and the following year the Toronto, Canada-based unit, with a facility in New Jersey, supplied the majority of explosives detection units required by the Transport Security Administration to enhance aviation security.

General

By means of HI-TIP, entire X-ray images of fictional baggage containing threat objects are automatically put between real baggage X-ray images during the inspection process. It is also possible to have single specific threat objects automatically projected into real X-ray images of scanned baggage.

The operator of the X-ray system is not aware of this projection process. While HI-TIP is active, the full range of image enhancement functions to support the evaluation process is available. Even automated operator support functions can be used in combination with HI-TIP. The task of the operator is to recognize and mark dangerous or suspect objects.

The Explosives detection section includes automatic high-speed X-ray systems for the non-intrusive inspection of baggage and other goods to detect predominantly explosives. This kind of X-ray equipment usually is an integral part of an airport's internal baggage handling system.

Millimetre-wave

Millimetre-waves lie in the spectral region between radio waves and infrared. This band possesses a unique property of passing transparently through lightweight materials such as clothing fabrics. The property is made use of in mm-wave imagers that find their principal application in concealed object detection systems – typically used to identify the presence of weapons and contraband that are hidden beneath a persons clothing.

Millimetre-wave imagers have evolved over the past 50 years from very rudimentary systems to today’s technologies that create live video-like images for use as advanced people screening portals. In this evolution from scientific instruments to commercial security devices, the overriding constraint has been the cost of the sensors that detect the mm-wave signals that are needed to produce an image.

To overcome this barrier, scanning systems have been employed to direct mm-wave energy from all parts of the scene onto a small number of sensors. Historically these scanning systems have been based on mechanical techniques employing moving parts to direct and focus the mm-wave energy.

Smiths use a revolutionary approach that scans the scene electronically and has resulted in the first ‘no moving parts’ imager that is suitable for mass people screening.   The eqo product – named because it senses mm-wave echoes – marks an evolution to ‘next-generation’ technology for mm-wave imagers. The electronic scanning not only dispenses with the need for moving parts but also dramatically reduces the footprint of the product. The system consists of a flat-panel array that produces excellent image quality with a real-time, full motion video style output.

Ion Mobility Spectrometry (IMS)

Ion Mobility Spectrometry is the science behind the IONSCAN products
Ion Mobility Spectrometry, or IMS, is an instrumental analytical method that is similar to Time-of-Flight MS. Instead of responding to molecular fragments, however, IMS uses soft ionization. Ionized molecules travel through a drift tube at distinct speeds that are related to their mass and geometry. Selectable positive and negative ionization enhances identification or sensitivity.

The ions in question are generated by atmospheric-pressure chemical ionization. Sample material is heated to yield a vapor that is swept into a small drift chamber where a beta radiation source ionizes the molecules. The resulting ions – separated according to size, mass and geometry – accelerate towards a detector.

On impact, each ion generates a specific signal, which is a function of ion mobility. Mobility (K) is determined from the drift velocity (vd) attained by ions in a weak electric field (E) in the drift tube, according to the equation, vd = K x E. The distribution of these signals forms an ion spectrum, with an ion mobility band corresponding to each of the unique ionic species. The spectrum is a fingerprint of the parent compound. Libraries of these ion spectra remain resident in the IONSCAN software and are in the screening process.

During analysis, the IONSCAN will look for ions that take the same amount of time and peak within an expected range of the target substances. If it finds ions that fit those parameters, it will detect the substance and “alarm’. If the IONSCAN does not find ions that fit those parameters, it will not alarm.

For more information on this technology, consult Ion Mobility Spectrometry by Gary Alan Eiceman and Zeev Karpas, published in 1994. This book covers theory, ionization processes, instrumentation, and qualitative and quantitative analyses. Our IONSCAN products embody these principles.

ETI

Echo Technologies Inc. (ETI) was established in 1997 as a sensor and instrumentation development company, specialising in techniques for the detection of harmful biological agents.  Its laboratory facilities are located near the Seaport District in Boston, Massachusetts. The company's initial business focus was directed toward the development of optical sensors and instrumentation for the detection of biological agents and chemical contaminants.
As well as protecting the public from the dissemination of biological and chemical warfare agents, it looked at public health threats, including new and emerging diseases, contaminated food products and poor indoor air quality. One example is the EchoSensor, a compact biological-warfare detection system, developed to monitor the presence of airborne bacterial agents.  ETI also adapted its core sensor technology for rapid detection of microbial contamination in drinking water and recreational waters. 
ETI has enjoyed collaboration with Harvard University, the University of British Columbia, Michigan Molecular Institute, University of South Carolina and the Army Research Laboratory on projects ranging from basic research to development of prototype sensor instrumentation.

The Bio-Seeq Portable Veterinary Diagnostics System provides onsite detection capability of pathogens in a wide range of environmental conditions. The system provides laboratory quality analysis in a portable field deployable format. Smiths Detection is a globally recognised leader in the deployment of lab quality technologies in formats suitable for field use by none experts. The Bio-Seeq Portable Veterinary Diagnostics System contains 5 of the combined PCR/Sample preparation stations and allows the user to run the same assay on 5 samples or 5 different assays at the same time. All actions are then controlled by the instrument and a positive or negative result is displayed to the user at the end of the analysis.

Meteorological measuring

The purpose of meteorological (MET) measuring is to provide the warfighter timely and accurate environmental conditions occurring throughout battlespace.  This MET information enables comprehensive situational awareness of the atmospheric conditions that will impact weapons system selections, tactics, and targeting solutions.

MET systems collect meteorological data from both on-scene sensors and via reachback to DoD Numerical Weather Centers.  On-scene sensors include upper air balloon soundings, local surface conditions, and satellites.  An on-scene Mesoscale MET Model assimilates the collected surface and upper air measurements and produces a data cube of environmental conditions throughout the battlefield.  Using this MET “nowcast” model, MET Messages, MET Warnings, and tactical decision aids can then be accurately produced from the Mesoscale MET Model.

Modern Systems use GPS navigation and other technologies to increase accuracy.  Computer systems calculate weapon and ammunition data needed for accurate shooting solutions.

Systems currently under development will provide continuous MET observations, satellite imagery, Doppler Radar, and Numerical Weather Products.  System size is being reduced while incorporating state-of-the-art technology.  The next generation of systems will be used by a wider array of military forces.

Color assignment

The scale comprises colors of orange for elements of low atomic number, i.e. elements which can be found in organic material, green for elements of medium atomic number and blue for elements of higher atomic number. The information concerning material thickness respectively absorption factor is furnished by means of the degree of brightness for the color signal. Therefore, items composed of the same material but which are different in thickness, show the same color while differing in brightness.

LATE-PCR

LATE (Linear-After-The-Exponential)-PCR, is an advanced form of asymmetric PCR developed by the Lawrence Wangh Laboratory at Brandeis University in Waltham, MA. In 2005, Smiths entered into an exclusive license agreement with Brandeis University for LATE-PCR and related technologies.. The agreement included a co-development of LATE-PCR funded by Smiths resulting in a number of further patented technologies that incorporate LATE-PCR, all of which are exclusively licenced to Smiths.

The basic principle of LATE-PCR involves the amplification of only one side of a DNA duplex, resulting in a high proportion of single strandeded DNA, which allows probing over a much wider temperature range (and thus a greater degree of mutlpiexing) than is possible with symmetric PCR.

The use of single stranded DNA amplicons provides a number of other advantages:
• Separation of Annealing and Detection (faster thermocycling)
• Larger Detection “Temperature Space” (more multiplexed assays)
• Easier to Design Low-Tm Probes (more multiplexing and faster thermocycling)
• Substantial Increase in Allele Discrimination (better integrity of amplification)
• Substantial Decrease in Background Fluorescence (better sensitivity)
• Probe/Target Saturation - Higher Signal, no Inhibition of Amplification (faster thermocycling and better sensitivity)
• Linear Kinetics with little scatter among replicates providing Quantitative Endpoint Analysis 

LATE-PCR reduces mispriming while maintaining amplification efficiency.

LATE-PCR is further enhanced by additional technologies that improve sample preparation, suppress amplification errors, improve probe design for rapid high-resolution analysis of the amplified product, make multiplexing easier, allow for rapid DNA sequencing, and enhances data analysis.

Bio-Seeq Sample Preparation Unit (SPU)

The Bio-Seeq SPU accepts a wide range of solid and liquid samples including blood, vesicular tissue, swabs etc. It includes all the chemicals needed to extract and purify DNA or RNA and can be stored at room temperature.
Once the sample is introduced, no further operator intervention is required.

Overlapping material

The advantage of a continuous color scale becomes evident when considering overlapping materials: In systems with abrupt color switch-over between organic and non-organic materials, even thin layers of overlapping materials, such as steel, copper or PVC, will lead to organic materials being classified as non-organic materials, which is wrong.


HI-MAT Plus, however, provides a mixed color representation depending on the degree of the mixture of materials which can be distinguished from the color of the overlapping materials.

Operation

Suspicious material that in the past has been difficult to recognize by the operator of standard baggage inspection units are automatically marked, i.e. without any further operator manipulation. The frames are color-coded:


  • Red frame: Explosives
  • Green frame: Narcotics (option)
  • Blue frame: High density (option)

The Check-In applications (airports) section includes X-ray systems for use in the airport check-in area. These systems usually are integrated in counters to provide non-intrusive inspection of passengers' baggage during the check-in procedure. All X-ray systems in this category are based on the proven HiTraX technology platform, offering superior image quality and excellent usability.

Environmental Technologies Group (ETG)

Environmental Technologies Group (ETG), located in Edgewood, Maryland, USA, had a long history of developing and manufacturing chemical detection systems for the US military, foreign military customers, and commercial customers. It went on to develop the Bio Detector, the detector of choice for the U.S. Army, jointly developed and field-tested with the U.S. Army Chemical and Biological Defense Command.

ETG's origins stretched back over 100 years, originally as Friez Instruments, later becoming a division of the Bendix Corporation.  ETG eventually established a leading position in the supply of hand held nuclear, biological and chemical agent detectors for military defense and first responders.

Farran Technology

Farran Technology developed millimetre wave technologies for the detection of hidden weapons and explosives.

Farran was founded in 1977 in Cork, Ireland, by senior research staff from the nearby National MicroElectronics Research Centre – one of Europe’s largest micro-electronics research establishments. 

Farran Technology grew to become a primary supplier of leading-edge millimetre-wave subsystems to major systems integrators. The company built a world-class level of IPR, embodied in its own patented designs, in its exclusive access to patented products and in the know-how of its long serving technical staff. 

Following the acquisition by Smiths, Smiths Detection took on the development of mm-wave based imaging systems and the advanced Tadar portal.

The Farran components business serving the broadband wireless, radar, defence and satellite applications was moved onto the Smiths Interconnect division

IONSCAN 400B Course Description

The IonScan 400B is a highly sensitive, analytical instrument that uses Ion Mobility Spectrometry (IMS) technology to detect and accurately identify trace residues of a wide variety of narcotic and explosive substances. The equipment can be set up to identify either narcotics or explosives, but not both at the same time. For example, airports would typically use the IonScan 400B to search for traces of explosives where as detention facility may use the IonScan 400B to search for traces of narcotics.

The IonScan 400B Operators course will provide students with the skills to perform the daily operations and complete the general preventative maintenance for their IonScan 400B. The course will cover the following major topics:

  • Daily maintenance procedures
  • Start up and performing verification
  • Proper sample collection and swab analysis
  • Reacting to alarms
  • General maintenance
  • Powering down the equipment

During the IonScan 400B Operators course, each student will gain practical/hands-on experience in each of the topic areas listed above.

  • Prerequisites - None, this course is designed for the novice who has little to no experience with the IonScan 400B 
  • Target Audience - Operators of the IonScan 400B (ex. Security Personnel)
  • Duration 6-8 hours
  • Class Size Minimum of 4; maximum of 10

Bill Mawer – President
Bill received a Masters Degree in Electronic and Electrical Engineering from Birmingham University. After five years in the Army, he moved into the defence and aerospace industries.  He joined Smiths Aerospace as Director of Marketing and Strategy. Bill moved to Smiths Detection in April 2003, initially as President of Smiths Detection North America. In 2004 he became part of the headquarters team as Director of Strategy and Technology. 

Jay Lewington - Chief Technology Officer 
Jay has over 20 years experience in product development. In 1986, after a period of doctoral and post-doctoral research into the cloning of herbicide resistance genes, he joined Amersham International where he developed several molecular products.  Subsequently he played key roles in developing products for rapid microbiology testing, and for food hygiene & water purity.  In 1996 he joined AZUR Environmental, where he was responsible for R&D and Operations in the UK & US, including the development of the first fully automated toxicity testing system for water monitoring.  In 2000 Jay joined Smiths to establish a biology team developing military bio-detectors. He has since held several management positions, latterly global Director of Biological R&D, before the formation of the Diagnostics business..

Michael Dalton - Vice President of Sales & Marketing, Clinical
Michael has over 20 years of experience in sales, marketing and business development of medical device, and diagnostic products targeting laboratorians and physicians.  He has experience in building and managing a sales force and marketing organisation for innovative diagnostic products, including a successful career with Siemens Diagnostics, building on his solid sales and marketing leadership and management skills.
 
Doug Barson - Financial Controller
Doug is a ACMA qualified accountant with over 30 year experience in the engineering industry.  He joined Smiths in 1997 as Finance Director of Smiths Detection – Watford and was promoted to the position of Finance Director for the Smiths Detection North America group in 2001 following the acquisition of two US Detection businesses. Subsequently he became Finance Director of the UK/International Military Division and later Controller for the Business Development and Technology group, with financial control responsibility for the Military & Americas businesses R&D budgets. 

Jason Betley, PhD - Director of Product Development
Jason received a double Honours Degree in Chemistry and Biochemistry from the University of Southampton followed by a PhD in Synthetic Organic Chemistry and Immunology at the University of Sheffield. He was a Professional Research Associate at the University of Colorado and Hewlett Packard Palo Alto in DNA Microarray Synthesis from 1997; Chemistry and Biochemistry Manager at Adprotech Ltd from 1999; and R&D Manager at Prometic Biosciences Ltd from 2003. He joined Smiths Detection in 2006. Jason has 6 peer reviewed publications and is the inventor on 12 patent applications and granted patents. He is Resource Manager for all private venture and funded projects in Smiths Detection Diagnostics.

Carmelo Volpe, PhD - Director of Sales & Marketing, Veterinary
Carmelo graduated in 1991 with a degree in Biotechnology from DeMonfort University in Leicester. He went on to study the regulation of lipid metabolism at the genetic level at the University of Wales, Collage of Cardiff and obtained a Molecular Biology Ph.D. in 1995. During this time he identified a novel regulatory protein which later was identified as STAT 5. He joined the Karolinska Institute in Sweden to run a gene therapy project, and moved to Applied Biosystems in 2001 as a professional services consultant, before taking a senior commercial/business development role. He joined Smiths Detection in 2006 as the Business Development Manager for the veterinary diagnostics business.

John Czajka, PhD - Director of Business Development
John received a Ph.D., with a focus on Molecular Diagnostics, from Cornell University in 1997 and also holds an MBA in technology management (2006).  He joined Smiths Detection in 2003 as a Principal Investigator for PCR assay development, becoming Deputy Director of Bio R&D in early 2005, then Vice President of Technology Acquisition in August 2006.

Peter Mula -  Director of Operations and Supply Chain
Peter has 20 years in Biotech and Pharmaceutical operational and supply chain management, beginning with a start-up biotech instrumentation company that was purchased by Amoco Oil. At Amoco he was part of the team that created a successful spin-off that became a world leader in molecular genetics for use in disease management. In 2001 he moved with the business to Abbott Labs where he held management positions in supply chain and strategic purchasing for both their diagnostics and pharmaceutical divisions. Peter joined Smiths Detection in June 2008.

Amy Rocklin, JD, PhD
Amy is the senior Intellectual Property Counsel for Smiths Group plc. She is responsible for all aspects of intellectual property portfolio management, intellectual property transactions, promulgating and implementing policies and procedures and training for Smiths Group.  Prior to joining Smiths, she was in private practice at Foley & Larder LLP and Banner & Witcoff Ltd. representing clients from small start ups to large publicly traded companies.  Amy holds a J.D. from the George Washington University School of Law and a Ph.D. from the Department of Biochemistry, Molecular Biology and Biophysics at the University of Minnesota.

IONSCAN-LS Training Opportunities

Smiths Detection offers regularly scheduled operator training classes at our Danbury, CT facility. Below you will find class dates as well as the training agenda and directions to our office.  The IONSCAN-LS Operator course will provide students with the skills to use the IONSCAN-LS, as well as perform the necessary preventive maintenance for the unit. There is also an introduction to Method Development as related to the
IONSCAN-LS. 

Class size is limited to 6 students and requires a minimum of four (4) students. Classes that do not meet the minimum are subject to cancellation three weeks prior to training date.

Hyperspectral Imaging

Infrared spectroscopy with FTIR is the proven technique to identify molecules at ambient temperature.  The hyperspectral imaging technology combines the powerful spectroscopy to each pixel of an image.  The longwave infrared hyperspectral imager delivers an image of a scene where the “fingerprint” of the molecules is used to locate clouds of various chemicals without the need for illumination.  The longwave infrared radiation travels through the atmosphere allowing the detection at standoff distance up to several kilometres.  
Hyperspectral data has become the backbone of many chemical and biological detection applications.  Advancements in infrared detector focal plane arrays combined with expertise in spectroscopy applications have spawned a new breed of hyperspectral imaging instruments.  Applications for such instruments are wide, from microscopic spectroscopy in medical labs to mineralogy studies on extraterrestrial sites to early warning systems for chemical agent and toxic chemical detection.  Telops has developed an instrument which provides an early warning for chemical agents and toxic chemicals, and also provides a “Chemical Map” of the field of view while being man portable.
Advancements in Mercury Cadmium Telluride focal plane arrays (FPAs) in recent years have allowed high performance longwave infrared imagers to prosper.  In particular, gas and chemical spectroscopy applications are being vastly improved with these new technologies.  A new breed of instruments, called hyperspectral imagers, has been introduced that provide three dimensions (two spatial and one spectral) of information per “image”.  However, for the transition from traditionally single pixel spectrometers to FPA based hyperspectral imagers to succeed, a couple of parallel advancements had to be made as well.
To provide the best field imaging spectroscopy instrument, Telops has developed the FIRST, Field-portable Imaging Radiometric Spectrometer Technology, instrument.  This instrument is based on a modular design that includes: a high performance infrared FPA and data acquisition electronics, onboard data processing electronics, a high performance Fourier transform modulator, dual integrated radiometric calibration targets, and a visible boresight camera.  These modules, assembled together in an environmentally robust structure, used in combination with Telops’ proven radiometric and spectral calibration algorithms make this instrument a world-class passive standoff detection system for chemical imaging.

Advanced Infrared Sensors
Telops' advanced infrared sensors provide radiance characteristics of targets in four dimensions, spectrally, spatially (2)  and temporally.  They make it easier to acquire the critical information about your targets.  Information such as, “Is there a gas present? Where is it?”, “Is there a hidden vehicle nearby?” and “How is the signature evolving?”
The Field-portable Imaging Radiometric Spectrometer Technology, the FIRST sensor, is optimised to provide unparalleled spatial information about the spectral characteristics of infrared targets.  With the FIRST, you can study the spatial distribution of the spectral signature of your targets in more than 80,000 simultaneous field of views.

Fourier-Transform Infrared Spectroscopy

Fourier-Transform Infrared spectroscopy (FT-IR) is a well-established and proven analytical technique for the identification of unknown chemicals. The method relies on the microscopic interaction of infrared light with chemical matter via a process of absorption and results in a pattern of bands called a spectrum. This spectrum is unique to the chemical and acts like a “molecular fingerprint” in the same way a human’s fingerprint is distinct.

FT-IR exploits the intrinsic properties of a chemical, in addition to being broadly-applicable. These features make FT-IR very amenable to spectral library-matching. With the help of an extensive database the library-matching approach makes it possible to identify rapidly thousands of chemicals on the basis of their distinct “molecular fingerprint”.

Raman spectroscopy

A complementary technology to FT-IR, Raman spectroscopy is a technique that identifies unknown chemicals by recording how their uniquely vibrating molecular bonds scatter incident laser light into distinct frequencies.

The key advantage of Raman spectroscopy is its ability to penetrate a variety of glass and plastic containers such that the potentially dangerous unknown substance need not be handled directly.

As with FT-IR, extensive Raman libraries allow thousands of chemicals to be identified on the basis of their distinct molecular fingerprint.

Ergonomic image display

Due to sophisticated real-time image processing and edge enhancement of the new HiTraX X-ray technology, the image focus of the HI-MAT Plus function is superior. Very fine structures and differences in density become visible so that thin wires or hidden items in manipulated luggage, can easily be recognized.

The sensitivity of the human eye varies with the wavelength (color) of light. The color representation has therefore been adapted to the human eye anatomy.

Given the HI-MAT Plus method, the coloring of the different materials in the image is chosen so that the brightness of color perceived by the human eye matches the luminance of the corresponding b/w image. The adaptation of the image representation to the physiognomy of the eye facilitates the focusing and object perceptibility.

Smiths Detection Develops Diagnostic Platform For Infectious Diseases

London, UK – 4 November, 2008. Smiths Detection today announces that it is developing a rapid diagnostic system, initially for use in hospital screening programmes for infectious diseases such as MRSA and Clostridium difficile.

The Bio-Seeq system will provide easy-to-use tests suitable for either a laboratory or Point of Care setting where it can be operated by non-specialist staff. Clinical trials are expected to start early next year.

The technology platform, for which Smiths Detection expects to provide a wide range of tests, is based on the Bio- Seeq portable veterinary laboratory which is due to undergo field trials for Foot & Mouth Disease testing with the UN  Food and Agriculture Organization later this year. 

Using the clinical diagnostic version of Bio-Seeq, the patient sample is introduced into a disposable Sample Preparation Unit which is then placed on the instrument. No manual sample preparation steps are needed and the unit can process a variety of solid and liquid samples making it ideally suited for combating healthcare-associated infections which kill thousands of patients each year.

A Reagent Pack, inserted into the Sample Preparation Unit, contains all the components for the specific test and includes a bar code used to set up the instrument, further reducing the operator input required. It is anticipated that Reagent Packs for a wider range of tests will be developed, either by Smiths Detection or selected development partners. The tests will use the LATE PCR technique licensed from Brandeis University in Boston, Mass. to amplify and analyse the genetic material of the infectious agent. The power of LATE PCR will be used to detect multiple diseases in each test, and to provide clinically important information such as antibiotic resistance and genetic mutations.

Stephen Phipson, Group Managing Director of Smiths Detection, said: “Our ability to transform sophisticated laboratory technologies into reliable, easy-to-use detection systems is one of our key skills. Applying this to clinical diagnostics is a logical step for us.”

A blue frame is inserted where the material to be checked is very dense and the system can no longer discriminate due to the high X-ray absorption. X-ACT operates without interrupting other evaluation procedures (e.g. no stopping of conveyor belt necessary).

X-ACT marks the substances in the HI-SCAN images. This means operators, using our standard systems, are familiar with the images and have an additional indication concerning the a. m. objects located in the bag. All other standard image evaluation functions are still available.

Cyrano Sciences

Founded in 1997, Cyrano Sciences, Inc., commercialised a simple, accurate, non-invasive tool that enabled machines to smell. 

It developed the electronic nose, using chemical sensors to sniff out explosives, food contaminants, and other compounds and to verify solvents and other chemicals. Its technology was also capable of recognising by smell certain medical conditions.

Cyrano was the worldwide exclusive licensee of the patented nanocomposite sensor technology developed at the California Institute of Technology. 

The Cyranose 320, commercialised in 2000, was the first major product development, as an efficient, affordable way to perform on-site analysis accurately. 

Cyrano later developed its capability to produce low cost, low power array based sensors for distributed monitoring, personnel protection or consumer products.

Due to the high technology standard all HI-SCAN units are equipped with a lot of image evaluation and enhancement features which are available to the operator. The HI-MAT feature displays 3 groups of material in different colors, whereas the Superenhancement function serves to automatically select the best contrast for every spot within the X-ray image. Biological agents

Daylight saving time procedures

In the light of the U.S. Energy Policy Act, 2005 and the changes to start and end dates for Daylight Saving Time, some Smiths Detection products operating in North America may need manual adjustment to the system clock. We are pleased to provide information on this process for our products impacted by this change.

Military

Sensor Management Solutions - Smiths Detection provides enterprise sensor management solutions for mission critical applications, developing intrusion detection software for physical security applications. At the heart of our systems is FirstView, a powerful web application based software solution for managing your matrix of security systems, from cameras to sensors, including CCTV, Video Analytics, CBRNE, Access Control and X-ray. It functions easily and securely over any network for Command & Control or even mobile applications.

Missed objects

If the operator has missed an object projected on the image, the conveyor automatically stops and a message is displayed on the screen.

Real X-ray images and missed threat object images are automatically stored. These images can be recalled using the IMS Image Store System.

Container screening with nuclear detection

To help authorities in the fight against dirty bombs and illegal imports of radioactive sources, Smiths Detection offers automatic radioactive material detection as an option on all HCV cargo screening systems - Gamma and/or neutron detection. The solution enables multi-threat detection with a single scan, screen and operator respectively.

The integration of radioactive material detection in the HCV systems is an innovative enhancement which allows the system to detect both illegal merchandise and radioactive materials located inside truck and container loads, without them having to be opened.

Chemical detection - Air systems

Bio-Seeq Plus - Handheld, fully field portable PCR identifier to detect bacterial and viral pathogens in 60 minutes.

The result: the image evaluation of materials that are difficult to penetrate is improved without deteriorating the information of other image sections.
HI-SPOT is an automatic function. Manual manipulation or stopping of the conveyor is not necessary.

HI-SPOT works online and in real time without a loss of evaluation time for the operative. Since the absorption threshold and the minimum size of the detection area are programmable parameters, HI-SPOT can be easily adjusted  by supervisor personnel.

GID-2A - fixed Chemical Agent Detector for continuous detection, for integration into collective protection systems, at sea or on land.

Operation

If a suspect object has been detected and marked by the operator, the conveyor automatically stops. The fictional threat object is now displayed with a black-and-yellow frame.

The fictional object disappears from the screen and the further evaluation of real screened baggage continues.

Decontamination

Explosives detection

We use two principal technologies to help you detect the deadly threat from explosive substances.

Trace detection products include hand-held and table top equipment to analyse trace particles on baggage or clothing and a walk-through portal to screen people.

X-ray systems for checked baggage that can be integrated into baggage handling or multi-level screening systems, and the first system to allow explosives detection for carry-on baggage at security checkpoints, using independent multi-energy generators and state-of-the-art X-ray sensor technology.

Graseby Dynamics

Graseby Dynamics established a world leading position in products that detect potentially lethal chemical and biological warfare agents. It was at Graseby that scientists developed the Ion Mobility Spectrometry (IMS) technology that lies at the heart of many Smiths Detection trace detection systems. 

In the 1970s Graseby began supplying the UK Ministry Of Defence with products  for anti terrorist work, to detect explosive vapours and it became a world leader in vapour detection, based on IMS. That expertise was extended in the 1980s to chemical warfare agents for the MoD. 

A major advance into biological detection came in the late 1990s when Graseby won the prototype biological detection system (PBDS) for the UK MoD, as the forerunner of today’s bio detection and identification systems.

Graseby plc was acquired by Smiths in 1997 and Graseby Dynamics became the company's first venture into the detection sector.

Sample Preparation Unit

All the Smiths Detection Diagnostics systems have the benefit of a disposable preparation unit for samples, a breakthough in technology and processing.
The SPU includes all the chemicals needed to extract and purify DNA or RNA and can be stored at room temperature.
One cartridge accepts any sample, ensuring wide suitability.
Any test can be carried out on the one cartridge, giving total flexibility.
And there's no instrument set-up, which means all tests can be run by non-experts.

To conduct a test the user inserts a reagent pack for the suspected disease into the SPU. The sample is introduced and the SPU is placed on to a vacant station on the instrument. The user presses “start” on the touch screen and all sample preparation and analysis is carried out automatically.

The key advantage of the system: Once the sample is introduced, no further operator intervention is required.

Running a test on the SPU is simplicity itself.
